Arbuscular mycorrhizal fungi (AMF), as obligate symbionts, play a key role in enhancing nutrient acquisition, improving tolerance to biotic and abiotic stresses and promoting sustainable plant disease management. Despite the economic importance of Golestan Province rangelands and the dependency of wild Gramineaeon AMF, there remains a considerable knowledge gap regarding the identification of these fungi in rangeland ecosystems. Accordingly, the present study was aimed to morphologically identify AMF communities associated with the dominant wild Gramineaein the southern rangelands of Golestan Province. For this purpose, the sampling was conducted at a depth of 5-30 cm around the rhizosphere of blue barley (HordeumglaucumSteud.),poa or winter grass (Poaannua L.),rye grass (Lolium perenne L.), and woolly brome (BromustomentellusBoiss.) in the rangelands of Nodeh Khanduz, Aqband, Alang, and Kabudwal in early summer 2022. Root samples were stained using lactoglycerol blue solution. AMF spores were extracted using wet sieving and sucrose centrifugation techniques to confirm the presence of symbiotic associations. Using standard identification keys and authoritative international databases, fungal species identification was carried out based on spore morphological characteristics, including shape, color, size, surface ornamentation, spore wall structure, hyphal attachment, etc. Morphological assessments revealed 21 AMF species belonging to Glomus (G. aureum, G. ambisporum, G. macrocarpum and G. sinuosum), Rhizophagus (R. intraradices and R. irregularis), Funneliformis (F. mosseae and F. geosporum), Septoglomus (S. viscosum, S. turnauae and S. deserticola), Claroideoglomus (C. drummondii, C. etunicatum, C. walkeri, C. lamellosum and C. claroideum), Entrophospora (E. furrazolae), Acaulospora (A. bireticulata), Scutellospora (S. graeca) and Pacispora (P. franciscana and P. scintillans) were identified. The species S. graeca and E. furrazolae were reported for the first time from the Iranian fungal flora. Furthermore, host-specific analysis revealed that H. glaucum maintained the highest AMF diversity (12 species), while the rhizosphere of L. perenne exhibited the lowest lowest richness (4 species). Overall, the findings demonstrate that the identified native arbuscular mycorrhizal fungi possess significant potential as effective inoculants for enhancing the restoration efficiency of grass cover in degraded rangelands of the region.
