Cryopreservation of Dracocephalum kotschyi seeds using with vitrification and encapsulation-dehydration

Today, preservation of species diversity of medicine-rangeland and prevention of genetic erosion is important. Dracocephalum kotschyi Boiss is one of the most important plants that exposed to extinction due to the irregular removal and climate changes. Hence, in the present study, the effects of encapsulation-dehydration and vitrification with two plant vitrification solutions (PVS2 and PVS3) on Dracocephalum kotschyi Boiss seeds were studied. The seeds were frozen after treatment with PVS2 and PVS3 solutions with two capsule-dehydration and vitrification methods and kept for 20 hours in a liquid nitrogen tank at - 196 ° C. The seeds were then melted at 40 ° C in Bain Mari. The seeds were cultured in petri dishes and in a pot. The rates of germination, seedling vigor index, root length, stem length and fresh and dry weight of the plants were evaluated.  Encapsulation-dehydration significantly decreased the rates of germination index and seedling vigor index in comparison with vitrification. Minimum and maximum of seedling length was obtained in treatments with encapsulation-dehydration and vitrification respectively with dehydration solution of PVS3.  Root dry weight was not influenced by different treatments of cryopreservation. however, encapsulation-dehydration significantly decreased the dry weight of shoot. The results showed that the treatment of vitrification with a dehydration solution of PVS3 decreases the stresses of the transition to the conditions of cryopreservation. It can in turn improve  the seeds quality of Dracocephalum kotschyi .
